FIX&PERM Cell Fixation and Permeabilization Kit

Flow cytometric analyses with monoclonal antibodies were so far mainly restricted to cell surface molecules. Intracellular structures such as cytoplasmic or nuclear enzymes, oncoproteins, cytokines, immunoglobulins etc. were largely excluded from such studies. Also excluded from flow cytometric studies were cytoplasmic localizations of well-established membrane molecules like CD3 and CD22, which, in their cytoplasmic form, are the most reliable lineage markers in undifferentiated leukemia. With the FIX&PERM® Kit flow cytometric analysis of intracellular antigens has become as easy as surface antigen studies. The only prerequisite is the availability of suitable antibody conjugates. Most of the available monoclonal antibody conjugates can be used with the FIX&PERM® Kit, some determinants are sensitive, however, to the fixation step involved. This and the optimal fixation time have to be tested for each reagent.

We are pleased to announce that Peter Rutten has now started in his role as Operations Director here at Exalpha Biologicals Inc. Peter has a Master’s of Science degree in Industrial and Organizational Psychology and this has lead him down a very business orientated career path. Peter is looking forward to working with the team at Exalpha. Background

The metabotropic glutamate receptors (mGluRs) are key receptors in the modulation of excitatory synaptic transmission in the central nervous system. They are implicated in many forms of neural plasticity as well as learning and memory and drug abuse (Bhattacharya et al., 2004; Francesconi et al., 2004; Wilson and Nicoll, 2001). Group I metabotropic glutamate receptors (consisting of mGluR1 and mGluR5) are G-protein-coupled neurotransmitter receptors that are localized in the perisynaptic region of the postsynaptic membrane. When activated, Group I mGluRs lead to stimulation of phospholipase and activation of Protein Kinase C. In contrast activation of Group II metabotropic receptors (mGluR2 and mGluR3) leads to inhibition of adenylate cyclase. The mGluR1 receptor may also be critically involved in limiting the deleterious effects on excitoxicity (Blaabjerg et al., 2003). In contrast, the mGluR5 receptor appears to be essential for late phase LTP in area CA1 of the hippocampus (Francesconi et al., 2004).

Immunogen

Synthetic peptide derived from the C-terminal region of the mGluR5 and mGluR1a proteins.

Positive Control

HEK-293 cell lysate. Specificity: Specific for the ~125k monomer and the ~250k dimers of mGluR5 and mGluR1. Immunolabeling is blocked by preadsorption of antibody with the peptide used as antigen to generate the antibody.

Provided in HEPES (pH 7.5) solution containing 150 mM NaCl, 100 µg per ml BSA and 50% glycerol
