Background

The collagenases are members of the matrix metalloproteinase (MMPs) family and at least three distinct forms (MMP-1, MMP-8 and MMP-13) have been identified. Collagenases are produced by many myeloid and non-myeloid cell types. Importantly, collagenase production has been found to increase in inflammation and also in tumor metastasis. Although the collagenases degrade collagen types I, II and III, their substrate specificity varies depending upon their role in physisological and pathological turnover of collagen. This latent pro-collagenase can be used for various purposes such as a reference standard for assaying collagenase activity. The most critical process in assaying for collagenase is the activation of inactive collagenase, since this varies depending upon the method, reagents and type of enzyme. Thus, it is important to use the same reference standard for individual tests. Furthermore, it can be used as a control for testing collagenase inhibitors.
