Background
This outstanding recombinant Protein A is coupled through a cysteine at the carboxy terminus creating an ether linkage to highly cross-linked agarose. It contains 6 mg of recombinant Protein A per ml of beads. The combinantion provides superior performance, binding 50 mg of human IgG per ml of beads under static conditions. Binding under flow will be less depending on the flow rate for all Protein A and Protein G products. Flow rate of 30 to 300 cm/hr are possible with this highly cross-linked agarose product under gravity flow, but it is not designed to pump under high pressure.

Formulation
Provided as a 50% aqueous suspension of Protein A in 20% ethanol solution.

Applications
Ligand Density: approximately 6 mg Protein A per ml of drained gel Dynamic Binding Capacity: approximately 35 mg human IgG per ml of drained gel (estimated in a 0.7x5cm column at a linear flow rate of 300 cm/h under the following conditions: binding buffer: 0.1 M PBS pH 7.0, Eluting buffer 0.1 M citrate pH 3.0. 2.8 times excess IgG of the binding capacity was applied. Bead Structure: 4% highly cross-linked agarose Bead Size Range: 45 to 165 µm Mean Bead Size: approximately 90 µm Maximum Linear Flow Rate: >1300 cm/hour at 25 deg C in an XK 16/20 column, bed height 5 cm pH Stability: Long term storage - 3 to 9; short term storage - 2 to 10 Chemical Stability: IgG binding capacity and recovery maintained after storage for (a) 7 days at 40º C in 3M NaSCN, 6M guanidine-HCl, 0.1M glycine (pH3.0), 70% ethanol; 10 hours at 23º C in 8M urea, 0.1M glycine (pH 11), 5.8M acetic acid (pH 2.0) Physical stability: Negligible volume variation due to changes in pH or ionic strength Sanitization: Sanitize the packed column with 2% Hibitane/20% ethanol or with 70% ethanol
