Protein-Coupled Receptors (EDG/S1P/LPA Products)
EDG receptors
are a novel family of G protein coupled receptors for lysophospholipid
(LPL) mediators such as sphingosine-1-phosphate (SPP) and lysophosphatidic
acid (LPA). There are currently 8 members of this growing family of receptors
(EDG 1-8). The principal ligand for EDG-1, -3 and -5 is SPP and the ligand
for EDG-2, -4 and -7 is LPA, EDG-6 appears to be a low affinity receptor
for SPP. The rat EDG-8 receptor has 50% homology to human EDG-1, -3 and
-5.
EDG receptors
couple to multiple types of G proteins to signal through ras, MAP kinase,
Rho, phospholipase C and several protein-tyrosine kinases. EDG-3
and -5 are mediated by Gi and Rho via activation of ELK-1 and SRF transcription
factors. EDG-5 (also known as H218 or AGR16) may co-precipitate with ERAP
(EDG receptor associated protein) a protein with a PDZ (PSD-95, DLz, ZO-1)
like domain.
